Leptin Inhibits Insulin Secretion In ob/ob Mice

The receptor for leptin is expressed in pancreatic
islets of ob/ob mice, and leptin inhibits insulin secretion in these animals. Dr.
Michael A. Cawthorne reports in this month's issue of Diabetes that the finding
"...provides evidence that hyperleptinemia might be part of a link between obesity
and diabetes."

Dr. Cawthorne and other researchers at the University of Buckingham, U.K.,
detected mRNA encoding the full-length leptin receptor in pancreatic islet,
hypothalamus, liver, kidney, spleen and heart cells in ob/ob mice. The receptor is
also expressed in these cells in lean mice.

Exogenous leptin inhibited insulin secretion in hyperinsulinemic and
glucose-stimulated ob/ob mice, but not in db/db diabetic mice, which lack a
functional leptin receptor. The finding "...strongly suggests that these effects are
receptor mediated," according to Dr. Cawthorne.

"These data suggest that leptin overproduction, particularly in abdominal fat, may
modify insulin secretion directly and could be involved in the development of the
diabetic syndrome," the British team concludes.
